چکیده

In this study, cell death regulation and induction in AML line from a relapsed MLL-rearranged model (MOLM-13) was investigated with doxorubin (Dox) betulinic acid (BetA), singly combination. CyQUANT Direct® Annexin V/propidium iodide double staining were used to measure the cytotoxic effects of compounds, respectively. Reactive oxygen species (ROS) generation measured using 2′,7′-dichlorofluorescin diacetate staining. Expressions proteins genes examined by Western blot reverse transcription polymerase chain reaction analysis, BetA (20 μM) Dox (1 indicated synergistic growth inhibitory effect on MOLM-13 cells. The combined drug caused more cells reside irreversible late apoptotic stage compared single treatments (p < 0.05). Elevation ROS may be mechanism involved since can directly disrupt mitochondrial activity. contrast, leukaemic U-937 cells, combination attenuated Dox-induced death. selectively reduced 0.05) recently reported anti-apoptotic Bcl-2 protein isoform p15-20-Bcl-2 our group, without affecting usually p26-Bcl-2-α. Further studies known inhibitors apoptosis are required confirm potential Dox-BetA modulate these pathways.

Bax-induced apoptotic cell death.

A is an ordered cascade of enzymatic events that culminates in cell death and the cleavage of DNA into characteristic nucleosomal fragments. It is operative during embryonic development and, during adult life, plays a key role in various physiological processes, such as tissue remodeling and execution and regulation of the immune response. The consequences of inappropriate apoptotic responses a...

Importance of ROS-mediated autophagy in determining apoptotic cell death induced by physapubescin B

Physapubescin B, a steroidal compound extracted from the plant Physalis pubescens L. (Solanaceae), has been reported to possess anti-cancer potential, whereas the molecular mechanism remains elusive. In this study, we first demonstrated that physapubescin B induced autophagy in human cancer cells based on the evidence that physapubescin B increased lipidation of microtubule-associated protein 1...

SIRT3-SOD2-ROS pathway is involved in linalool-induced glioma cell apoptotic death.

Glioma is the most prevalent type of adult primary brain tumor and chemotherapy of glioma was limited by drug-resistance. Linalool is an acyclic monoterpene alcohol possessing various pharmacological activities. The present study was conducted to evaluate the effect of linalool on glioma cell growth. The effect of linalool on cell viability in U87-MG cells was investigated and the results showe...

Iron Overload Induced Apoptotic Cell Death in Isolated Rat Hepatocytes Mediated by Reactive Oxygen Species

Isolated rat hepatocytes in culture were incubated with different concentrations of iron-sorbitol (50, 100, 150, and 200 µM) to assess the changes in reactive oxygen species (ROS) and lipid peroxidation leading to apoptotic hepatocyte cell death. The viability of hepatocytes was declined depending on the iron concentration. One hour incubation of the cells with 100 µM iron resulted in decreased...
